This kit utilizes optimized detergent formulations to enable rapid and efficient isolation of nuclear and cytoplasmic proteins from animal cells. The nuclear fraction includes nuclear membrane proteins as well as soluble nuclear proteins, such as histones and transcription factors. The protocol reduces nuclear protein extraction time to approximately 10 minutes while delivering significantly higher yields compared to conventional extraction kits.
The system provides efficient nuclear-cytoplasmic separation, making it particularly suitable for Western Blot (WB) analysis.

Q1 : What are the functions of each component in BF7476 and BF7477?
A : The Reagent A induces swelling of the cell causing stress on the cellular membrane. The Reagent B reagent gently lyses the cell membrane allowing cytoplasmic proteins to be collected, while leaving the nucleus intact. The Reagent C is then used to extract nuclear proteins from the pellet.
Q2 : What is the difference between BF7476 and BF7477?
A : BF7476 contains detergent. The extracted nuclear proteins include nuclear membrane proteins, as well as histones and intranuclear transcription factors. It suitable for Western blot analysis.
BF7477 is detergent-free. It efficiently extracts soluble intranuclear proteins while excluding nuclear membrane proteins. After desalting or dilution, the isolated proteins are compatible with WB, IP, EMSA, Footprinting assays, reporter gene detection, and enzyme activity measurements.
Q3 : How can I minimize cross-contamination of proteins between the nuclear and cytoplasmic fractions when using our kits?
A : Careful pipetting and performing the incubation and centrifugation as indicated in the instructions will minimize cross-contamination. Performing an additional wash of the nuclear pellet with PBS before adding the Reagent C will reduce carryover of the cytoplasmic proteins to the nuclear protein fraction.
