This product enables the convenient and efficient separation of nuclear proteins and cytoplasmic proteins from animal cells or tissues. It gradually lyses cells, separates the intact nuclei from the cytoplasm, and then extracts nuclear proteins from the isolated nuclei.
Animal Nuclear and Cytoplasmic Extraction Kit (High-efficiency for WB) contains a strong detergent and can extract nuclear proteins, including not only nuclear membrane proteins but also histones and soluble proteins such as nuclear transcription factors. In traditional kits, the nuclear protein extraction time typically ranges from 30 to 40 minutes, whereas this product reduces the extraction time to just 10 minutes. Additionally, the amount of nuclear protein extracted with this product is significantly higher than that of other kits, and the nuclear-cytoplasmic separation is notably improved, making it especially suitable for Western Blot experiments.
Animal Nuclear and Cytoplasmic Extraction Kit (Multi-application Type) does not contain detergents and can effectively extract soluble proteins from the nucleus, but it does not include membrane proteins. After desalting or dilution, the separated nuclear and cytoplasmic proteins can be used in experiments such as Western Blot (WB), Immunoprecipitation (IP), Electrophoretic Mobility Shift Assay (EMSA), Footprinting, Reporter Gene Assays, and enzyme activity assays.

KEY FEATURES

High Purity

High Purity The nuclear protein/cytoplasmic protein separation is highly effective with an extremely low cross-contamination rate (better than traditional kits).

Convenient

Convenient Simple procedure with no need for gradient ultracentrifugation.

Rapid (High-efficiency for WB)

Rapid (High-efficiency for WB) Completed approximately one hour.

Unparalleled Yield (High-efficiency for WB)

Unparalleled Yield (High-efficiency for WB) This product is an efficient Western Blot reagent kit with an optimized formulation for nuclear protein extraction, yielding more protein than conventional protein extraction kits, making it ideal for Western Blot applications. Achieve up to 50% greater output.

High Yield (Multi-application Type)

High Yield (Multi-application Type) This product effectively extracts soluble nuclear proteins (excluding nuclear membrane proteins).

High Compatibility (Multi-application Type)

High Compatibility (Multi-application Type) The extracted product is suitable for experiments such as WB, IP, EMSA, Footprinting, reporter gene assays, and enzyme activity assays.

Fig 1. Protein was quantified using Ready-to-use BCA Protein Assay Kit (CAT. No. BH5484). Values are the average of two separate isolations.

The kit offers a high yield of protein, with 200-300 μg of cytoplasmic protein and 50-150 μg of nuclear protein at 1 mg/mL.

Fig 2. WB detection results of nuclear proteins from different kits. Equal amounts (2 × 10^6) of Expi293F cells were used for each kit. The extraction process was conducted following the manufacturer's instruction.

Compared to Thermo's kit, YamayBio BF7476 demonstrated a higher yield of nuclear proteins such as Histone H3, Histone H2A.X, and c-Fos, while maintaining comparable yields of cytoplasmic proteins.

Use two kinds of kits: YamayBio BF7476 and Thermo 78833 to extract nuclear proteins according to their respective instructions. WB experiments were conducted using two different antibodies.

Ensures minimal cross-contamination between nuclear and cytoplasmic proteins, with cross-contamination typically below 5%, providing high-quality nuclear protein extracts for downstream applications.
