Annexin V-FITC apoptosis detection kit is a kind of apoptosis detection kit, which uses FITC-labeled recombinant human Annexin V to detect phosphatidylserine on the surface of cell membrane during apoptosis. Flow cytometry, fluorescence microscope or other fluorescence detection equipment can be used for detection.
Cell apoptosis is one of the most common forms of cell death and is strictly regulated by genes. The process is often accompanied by significant changes in cell morphology. For example, in the early stages of apoptosis, phosphatidylserine (PS) normally located on the inner side of the plasma membrane flips to the outer surface. In the middle and late stages, as the cell membrane becomes more permeable, larger molecules like propidium iodide (PI), a DNA-binding dye, can freely diffuse into the cell and stain the nucleus with red fluorescence.
Annexin V is a calcium-dependent phospholipid-binding protein with a molecular weight of 35-36 kDa. It specifically binds to phosphatidylserine with high affinity. This assay kit uses Annexin V fused with green fluorescent protein (FITC) as a probe to detect phosphatidylserine. In combination with propidium iodide (PI), it enables the rapid detection of cell apoptosis using flow cytometry, fluorescence microscopy, or other fluorescence-based detection devices. After staining cell populations with Annexin V-FITC apoptosis detection reagent, cells undergoing early apoptosis exhibit green fluorescence, while cells undergoing late apoptosis and necrosis display both red and green fluorescence. Live cells show minimal fluorescence.
